Abstract:
ปัญหาโรคระบบทางเดินหายใจอันเนื่องมาจากการสูบบุหรี่ โดยเฉพาะโรคมะเร็งปอด ถือเป็นหนึ่งในปัญหาที่สำคัญของประเทศไทย โดยเอนไซม์ CYP2A6 ในตับที่ย่อยสลายนิโคตินก่อให้เกิดการเสพติดการสูบบุหรี่ในขณะที่เอนไซม์ CYP2A13 ที่ปอดกระตุ้นสารก่อมะเร็ง 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) ส่งผลให้เกิดโรคมะเร็งปอดขึ้น ดังนั้นการลดการทำงานของเอนไซม์ CYP2A6 และ CYP2A13 จึงเป็นเป้าหมายในการรักษาอาการเสพติดบุหรี่และลดโอกาสเสี่ยงในการเกิดโรคมะเร็งปอด โครงงานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาความจำเพาะและกลไกในการยับยั้งการทำงานของเอนไซม์ CYP2A6 ของสารสกัดจากสมุนไพรขลู่และหญ้าดอกขาว ที่เคยมีรายงานว่าออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 ได้ดีในหลอดทดลอง ผลการศึกษาพบว่าสารสำคัญในกลุ่มฟลาโวนอยด์จากพืชทั้งสองชนิดได้แก่ apigenin luteolin chrysoeriol และ quercetin ออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 และ CYP2A13 ได้ดีกว่าสารสำคัญในกลุ่ม thiophene ทั้งสามชนิดที่ทำบริสุทธิ์ได้จากขลู่ และสารบริสุทธิ์ในกลุ่ม hirsutinolide-type sesquiterpene lactones จากสมุนไพรหญ้าดอกขาวออกฤทธิ๋ยับยั้งได้น้อยที่สุด อย่างไรก็ตามสารในกลุ่มฟลาโวนอยด์ 4 ชนิด ออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 และ CYP2A13 แบบผันกลับได้ ในขณะที่สารในกลุ่ม thiophene และ hirsutinolide-type sesquiterpene lactones ออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 และ CYP2A13 แบบผันกลับไม่ได้ที่ขึ้นอยู่กับสารให้อิเล็กตรอน NADPH เวลาที่ใช้ในการบ่มสารยับยั้งและความเข้มข้นของสารยับยั้ง (Mecahinism based inhibition)
