Abstract:
ปัญหาโรคระบบทางเดินหายใจอันเนื่องมาจากการสูบบุหรี่ โดยเฉพาะโรคมะเร็งปอด ถือเป็นหนึ่งในปัญหาที่สำคัญของประเทศไทย โดยเอนไซม์ CYP2A6 ที่ตับย่อยสลายนิโคตินก่อให้เกิดการเสพติดการสูบบุหรี่ในขณะที่เอนไซม์ CYP2A13 ที่ปอดกระตุ้นสารก่อมะเร็ง 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) ส่งผลให้เกิดโรคมะเร็งปอดขึ้น ดังนั้นการลดการทำงานของเอนไซม์ CYP2A6 และ
CYP2A13 น่าจะช่วยรักษาอาการเสพติดบุหรี่และช่วยลดโอกาสเสี่ยงในการเกิดโรคมะเร็งปอดได้ โครงงานวิจัยนี้มีวัตถุประสงค์เพื่อทำบริสุทธิ์และศึกษาฤทธิ์ของสารสำคัญพืชในวงศ์ Asteraceae ได้แก่ ขลู่ ดอกคำฝอยและหญ้าดอกขาว และในวงศ์ Acanthaceae ได้แก่ ทองพันชั่งที่เคยมีรายงานว่าออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 ได้ดีในหลอดทดลอง ผลการศึกษาพบว่าส่วนสกัดเอทิลอะซิเตทของขลู่ออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 ได้ดีที่สุดและส่วนสกัดดอกคำฝอยเฮกเซนและเอทิลอะซิเทตออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 ได้ดีโดยส่วนสกัดย่อยเฮกเซนที่ 4 5 และ 7 ออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 ได้ดีมาก ผลการศึกษาเพิ่มเติมของสมุนไพรทองพันชั่ง พบว่าทั้งส่วนสกัดเอกเซนและเอทิลอะซิเทตออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 และ CYP2A13 ได้ดี โดยสารสำคัญจากทองพันชั่งที่ผ่านการทำบริสุทธิ์ได้แก่ Rhinacanthin-A,-B, และ-C ออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 และ CYP2A13 ได้ดี ด้วยกลไกการยับยั้งแบบผันกลับไม่ได้ที่ขึ้นอยู่กับสารให้อิเล็กตรอน NADPH เวลาที่ใช้ในการบ่มสารยับยั้งและความเข้มข้นของสารยับยั้ง (Mecahinism based inhibition) ต่อทั้งเอนไซม์ CYP2A6 และ CYP2A13 นอกจากนี้สารบริสุทธิ์จากสมุนไพรหญ้าดอกขาวอีก 8 ชนิดยังพบว่าออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 ได้ดี โดยที่สารในกลุ่มฟลาโวนอยด์ 4 ชนิดออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 แบบผันกลับได้ ในขณะที่สารในกลุ่ม hirsutinolide-type sesquiterpene lactones ออกฤทธิ์ยับยั้งเอนไซม์ CYP2A6 แบบผันกลับไม่ได้ที่ขึ้นอยู่กับสารให้อิเล็กตรอน NADPH เวลาที่ใช้ในการบ่มสารยับยั้งและความเข้มข้นของสารยับยั้ง (Mecahinism based inhibition)
Cigarette smoking constitutes one of the main causes of tobacco-associated respiratory diseases including lung cancer. The human cytochrome P450 CYP2A6 and CYP2A13 are
responsible for nicotine metabolisms and etabolic activation of tobacco-specific nitrosamines, 4-(methyl-nitrosamino)-1-(3-pyridyl)-1-butanone (NNK), generating metabolites that associated with lung diseases and cancers, respectively. Thus, inhibition of these two enzymes by specific inhibitor could an aid in smoking cession and lung cancer prevention. This study aims to characterize the active constituents and it’s inhibitory against activities in medicinal plants from family Asteraceae (Phuchea indica, Carthamus tinctorius, Vernonia cinerea) and Acanthaceae (Rhinacanthus nasutus) that previously reported to inhibited CYP2A6 and CYP2A13 enzymes in vitro. The results indicated that the Ethyl acetate fraction of P.inidica could potently inhibited CYP2A6 enzyme. The Hexane and Ethyl acetate fraction of C. tinctorius could equally inhibited CYP2A6 enzyme in which the Hexane sub-fraction 4, 5, and 7 of C. tinctorius were potently inhibited CYP2A6 enzyme. Interstingly, both Hexane and Ethyl acetate fraction of R. nasutus could potently inhibited CYP2A6 and CYP2A13 enzymes. The purify active conpounds, Rhinacanthin-A, -B, and –C were successfully isolated and characterized as an NADPH-, time-, and concentration dependent inhibition an indicator of a suicide inhibition or mechanism based inhibition (MBI) mode against both CYP2A6 and CYP2A13 enzymes. Moreover, Eight active compounds that comprise inhibitory activity toward CYP2A6 using activity-guided assays were successfully isolated from V. cinerea. These compounds were three flavones, one flavonol, and four hirsutinolide-type sesquiterpene lactones. Flavonoids possessed strong inhibitory effect on CYP2A6 in reversible mode, while inhibition by hirsutinolides was mechanism-based (NADPH-, concentration-, and timedependence) and irreversible against CYP2A6 enzyme.
