| dc.description.abstract | คณะผู้วิจัยพยายามพัฒนาวิธีการตรวจสอบ Babesia bovis ให้มีความไวและความจำเพาะสูงและมีความสะดวกในการตรวจสอบเลือดตัวอย่างได้ปริมารมากในคราวเดียวกัน การศึกษาในครั้งนี้ได้ทำการเพิ่มจำนวนชิ้นส่วนของยีน Carbamoyl Phosphate Synthetase II และตรวจสอบดีเอ้นเอผลผลิตด้วยวิธี enzyme-linked immunosorbent assay (ELISA) พบว่าเทคนิค PCR-ELISA นี้สามารถตรวจสอบการติดเชื้อ B.bovis ได้ต่ำถึง 0.000144% parasitemia หรือ เท่ากับเม็ดเลือดแดงที่ติดเชื้อจำนวน 10 เซลล์ในเลือด 1 ไมโครลิตน การตรวจสแบการติดเชื้อในเลือดตัวอย่างจำนวน 53 ตัวอย่าง พบว่ามี 5 ตัวอย่างที่ให้ผลบวกกับการตรวจสอบโดยเทคนิค PCR-ELISA แต่ไม่มีเลือดตัวอย่างใดที่ตรวจพบเชื้อ B.bovis โดย การตรวจด้วยกล้องจุลทรรศน์ นอกจากนี้เทคนิค PCR-ELISA สามารถตรวจสอบการติดเชื้อ B.bovis ในโคทดลองได้เร็วกว่าการตรวจด้วยกล้องจุลทรรศน์ คือตั้งแต่วันที่ 4 หลังการฉีดเชื้อ | th_TH |

| dc.description.abstractalternative | In this work we developed a sufficiently sensitive and specific PCR-based assay for Babesia bovis. PCR product was detected by enzyme-linked immunosorbent assay (ELISA) to simplify the processing of large number of samples. The primer and probe sequences were derived from Carbamoyl Phosphate Synthetase II gene sequences. The assay detected down to 0.000144% parasitemia (10 infected erythrocytes per microliter of blood). By the PCR-ELISA method, five of fifty three blood samples showed positive reactions, while with microscopic examination of blood samples were detected B. bovis. In addition, the PCE-ELISA can detected since day 4 after infection, in experimental cow, before detection by microscopic examination (day 10 after infection). | th_TH |
